Two Studies A World Apart: Same Conclusion
According to cancercenter.org, researchers at the University of Hong Kong studied more than 60,000 patients who took PPIs to treat H. pylori. These bacteria, which live in the digestive system, can cause ulcers and increase the risk of stomach or oesophagal cancer. The Hong Kong study concluded that long-term PPI use more than doubled the risk of stomach cancer. A Swedish study of patients on PPI therapy drew similar conclusions. But other research has contradicted those findings,
The health body Rare Diseases explains that stomach cancer arises from the cells lining the stomach, which like all cancerous cells, exhibit abnormal and rapid growth. In fact, stomach cancer was recently listed by the Less Survival Cancer Taskforce as rapid-growing cancer, alongside cancers of the lung, liver, brain, oesophageal, and pancreas.

Warnings For People With Certain Health Conditions
For people with osteoporosis: Pantoprazole can increase a persons risk for osteoporosis, a condition that causes bones to become brittle. Tell your doctor if you have a history of osteoporosis.
For people with low blood magnesium : Pantoprazole can decrease the amount of magnesium in your body. Tell your doctor if you have a history of hypomagnesemia.
For people being tested for neuroendocrine tumors: Pantoprazole can cause incorrect results in these tests. For this reason, your doctor will have you stop taking this drug at least 14 days before you have this testing. They may also have you repeat the testing if needed.
What Else Can I Do To Minimize My Risk Of Getting Stomach Cancer
Stomach cancer, as indicated by WebMD, is third in line for top global killer diseases. While a relationship between PPIs has been established, a direct connection has not been. There is a lack of clarity about the significance of the common H. Pylori infection. While many whose bacterial infections in the study were cleared up with antibiotics and who were maintained for two years afterward on PPIs still got stomach cancer, many did not.
This information should point the patient in the direction of making sure they speak with their physician about H. pylori screening and treatment if they experience frequent heartburn.
Other lifestyle changes to manage heartburn recommended by New York University Langone Health include eating six small meals rather than three larger meals throughout the day, keep your head elevated while you sleep by using an additional pillow or two, and avoid indulging in trigger foods as part of your diet.
Trigger foods are those that are high in caffeine, acids, oils, and spice. Examples include coffeeparticularly on an empty stomach, tomato-based products like spaghetti and pizza, fast deep-fried foods like hamburgers and French fries, and ethnic foods where hot condiments are used like Indian or Mexican dishes.

Diet & Lifestyle Advise
- Avoid intake of acid or heartburn triggering foods or drinks like onions, peppermint, chocolate, caffeinated beverages, citrus fruits or juices, tomatoes and high-fat and spicy foods.
- Before going to sleep try to raise your bedhead so that your head and chest are higher than your feet. Do not use piles of pillows, instead, one raised block is fine. This will not allow the stomach acid to backflow through your food pipe.
- Avoid taking alcohol and smoking cigarette. Alcohol can raise the level of production of stomach acid leading to heartburn and acid reflux. On the other hand, nicotine smoking damages the valve which prevents backflow of the stomach acid back into the food pipe.
- Include high fibre containing foods, berries, cherries, leafy green veggies and black peppers in your meal. These foods are full of antioxidant, calcium and vitamin B 12 that can help cope with the long term effects of the medicine. Fermented dairy products like miso, sauerkraut, and kimchi contain probiotics which help in the prevention of excess stomach acid production. Cranberry juice can be beneficial in the peptic ulcer and H Pyroli infection.
- Avoid regular sitting continuously, as it can increase stomach acid production. Try to take a break of 5 minutes in 1 hour by brisk walking or stretching.
Acid Reflux Drug Linked To More Than Doubled Risk Of Stomach Cancer Study
There are more than 50m prescriptions for proton pump inhibitors in the UK, though they have previously been linked to side-effects and increased risk of death
A drug commonly used to treat acid reflux is linked to a more than doubled risk of developing stomach cancer, researchers have claimed.
Proton pump inhibitors reduce the amount of acid made by the stomach and are used to treat acid reflux and stomach ulcers.
A study published in the journal Gut identified an association between long-term use of the drug and a 2.4 times higher risk of developing stomach cancer. In the UK, there are more than 50m prescriptions for PPIs every year but they have been linked to side-effects and an increased risk of death.
A link between PPIs and a higher stomach cancer risk has previously been identified by academics but never in a study that first eliminates a type of bacteria suspected of fuelling the illnesss development.
Research by the University of Hong Kong and University College London found that after the Helicobacter pylori was removed, the risk of developing the disease still rose in line with the dose and duration of PPI treatment.
They compared the use of PPI against another drug which limits acid production known as H2 blockers in 63,397 adults. The participants selected had been treated with triple therapy, which combines PPI and antibiotics to kill off the H pylori bacteria over a week, between 2003 and 2012.

Try This Heartburn Medication Instead
The good news is H2 blockers, another class of heartburn medications such as Pepcid and Tagamet , are safer alternative medications to the PPIs. The same study using H2 blocks as control medications to the PPIs showed that the stomach cancer risk of PPIs is 2-8 times higher than the H2 blockers.
Interestingly, the Presidents health summary published by The White House on November 19, 2021, revealed that the H2 blocker drug Pepcid was chosen by The White House physician to treat Presidents heartburn symptoms and acid reflux disease. This decision may be because this drug has not yet been linked with reported cancer risk.
Every medication has indications and side effects, including the H2 blockers Pepcid and Tagamet. You should consult with your doctors to choose the appropriate medication to treat your heartburn and acid reflex based on your conditions.
Prevention is better than treatment. To avoid heartburn, you should:
- eat smaller and frequent meals
- avoid late-night eating and
- not exercise right after meals
We hope these heartburn-smart approaches could prevent or reduce your heartburns during this holiday season.

Pantoprazole Enhances The Cytotoxicity Of Vitamin C And Increases Intracellular Ros Accumulation In Prostate Cancer Cells In A Ph
As determined by the WST-8 assay, in cell culture medium with an acidic pH , the proliferation of most cancer cells except OVCAR3 cells was unaffected by the combination of vitamin C and pantoprazole when the compounds were administered simultaneously . However, when cells were pretreated with pantoprazole for 24 h, pantoprazole single treatment caused a reduction in the viability of PC3 prostate cancer cells to 0.4 units and in DU145 prostate cancer cells to 0.7 units . Moreover, the reduction in cell viability was slightly more robust following combined administration of vitamin C and pantoprazole in both prostate cancer cell lines . In contrast, at an alkaline pH , both vitamin C single treatment and in combination with pantoprazole resulted in significant reduction of the viability in PC3 and DU145 cells. Compared to no pretreatment, pretreatment with pantoprazole followed by combined administration of vitamin C and pantoprazole caused an additional reduction in the viability of prostate cancer cells . Similar results were obtained for MCF7 and SKBR3 and SKOV3 cells. OVCAR3 showed somewhat different results .

Potential Chemopreventive Agents To Reduce Proton
Meta-analyses have shown that aspirin reduces gastric cancer risk without stratification according to H. pylori status., To address this issue, we conducted another study on the effect of aspirin specifically on H. pylori-eradicated subjects. Aspirin was shown to be associated with a lower gastric cancer risk among H. pylori-eradicated subjects , with a frequency, duration, and doseresponse relationship being observed. A subsequent post hoc analysis showed that the harmful effect of PPIs was higher among nonaspirin users, while PPIs were not associated with an increased risk of gastric cancer among aspirin users . The potential detrimental effect of PPIs on gastric cancer appears to be negated by aspirin use, although this observation necessitates validation by studies from other centers. Nevertheless, our study finding prompts the investigation of the role of other potential chemopreventive agents, such as metformin and statins, in reducing gastric cancer risk among PPI users should be further explored.

Strengths And Limitations Of This Study
This study has several strengths. First, to our knowledge, this is the largest study with the longest follow-up period conducted to date. Given the number of gastric events observed in our cohort, this study was sufficiently powered to address the long-term safety of PPIs and assess the risk among important subgroups, including by duration of use. Second, we restricted the cohort to new drug users, eliminating biases associated with the inclusion of prevalent users. Third, the comparator group consisted of patients prescribed H2RAs, an active comparator that likely minimised confounding by indication. Moreover, the use of propensity score-weighted methods ensured an excellent balance of all baseline confounders. Finally, our results remained highly consistent across several sensitivity analyses.
In summary, the results of this large real-world study suggest that patients newly treated with PPIs may be at an increased risk of gastric cancer compared with patients newly treated with H2RAs, although the absolute risk remains low. While PPIs have established clinical benefits when used according to evidence-based guidelines, this study highlights the need for physicians to regularly reassess the necessity of ongoing treatment. This is especially important in patients who are prescribed PPIs in the long term and for patients without an evidence-based indication for use.
Effect Of Pantoprazole On Human Gastric Adenocarcinoma Sgc7901 Cells Through Regulation Of Phospholrp6 Expression In Wnt/
This article is mentioned in:
Gastric cancer is the leading cause ofcancer-related mortality in China and is the third leading cause ofcancer-related mortality in North America and Western Europe. Previous studies have foundthat an acidic tumor microenvironment is key to managing cancerprogression and metastasis .Vacuolar-ATPases , specific proton pumps of the cell, have an important role in maintaining a relatively neutralintracellular pH , an acidic luminal pH, and an acidicextracellular pH . They are overexpressed in many types ofmetastatic cancers and are positively correlated with invasive andmetastatic tumor potential .Furthermore, blocking the expression of V-ATPases can inhibit thegrowth and metastasis of human cancer .
Materials and methods
Pantoprazole was purchased from Altana Pharma. According to our previous study, aconcentration > 20 mg/ml of pantoprazole inhibited theproliferation of SGC7901 cells. Pantoprazole was resuspended innormal saline at 20 mg/ml immediately before use.
